C2 antigen is measured by radial immunodiffusion (RID). Antiserum to C2 is embedded in a layer of agarose, wells are punched into the agarose gel, and the patient specimens and standards are allowed to diffuse into the gel and form a precipitin ring. The diameter of the ring is proportional to the antigen (C2) concentration.
