Centrifuged urine is mixed with a dimethylmethylene blue (DMB)/Tris hydroxymethlyaminomethane (TRIS) solution. The metachromatic dye DMB binds with glycosaminoglycans (GAG) and the complex formed changes the spectra of the dye.
