Specimen Required

Collect specimen as follows:

Note:   Draw on wet ice. Use refrigerated centrifuge

            (5 degree C) to spin. Freeze plasma within 20 minutes

            of collection.

 

1. Obtain venous blood by clean venipuncture. Avoid slow flowing

     draws and/or traumatic venipunctures as either of these may result

     in an activated or clotted specimen. Do not use needles smaller than

     23 gauge.

2. Always draw a pilot tube (7-10 mL red top tube, completely filled)

     before drawing coagulation specimens in light blue vacuum tubes

     (3.2% or 3.8% buffered sodium citrate). Discard the pilot tube.

Note:   Reference ranges have been established using 3.2%

                  buffered sodium citrate.

3. Fill light blue top tubes as far as vacuum will allow and mix by gentle

     inversion. Exact ratio of 9 parts blood to 1 part anticoagulant must be

     maintained.

4. Centrifuge the specimen at 1700 x g for 15 minutes (or at a speed and

     time required to consistently produce platelet-poor plasma - platelet

     count less than 10,000/uL. Hemolyzed specimens will be rejected.

5. Immediately remove only the top two thirds of the platelet-poor

     plasma from the specimen using a plastic transfer pipet (use of glass

     transfer pipets may result in activation and/or clotting of the

     plasma). Place the plasma in a properly labeled plastic vial and

     clearly mark the vial contents as PLASMA. GLASS VIALS WILL BE

     REJECTED.

6.  Quick freeze the plasma specimens using a dry ice and

     methanol bath or in a -60 degree C to -70 degree C freezer. Each

     assay requested must be submitted in a separate plastic vial.

7. Send 2.0 mL of platelet-poor plasma (sodium citrate) FROZEN

     on dry ice.

Note:    1. Each coagulation assay requested should have

                  its own separate aliquot of plasma.

                  2. Specimens containing heparin should not be used

                  for coagulation studies. If possible, stop heparin

                  therapy before the draw to avoid contamination.

                  Heparin interferes with most clotting assays.

                  3. Include a brief patient history and other pertinent

                  clinical information (i.e. medications, blood

                  products, etc).
