Genomic DNA is extracted from whole blood. The portion of the ACVRL1 gene containing the identified familial mutation is amplified and sequenced in both directions using florescent dye-terminator chemistry. Sequencing products are separated on an automated sequencer and trace files analyzed by mutation detection software and visual inspection for the familial mutation in the exon of interest.(Unpublished Mayo method)
