The detection of sex chromosome mosaicism is based on FISH analysis of the centromere regions of the X and Y chromosome. A total of 500 interphase nuclei (250 each by 2 technologists) from peripheral blood cultures are examined for the presence of the X centromere (green signal) and the Y centromere (orange signal).(Wiktor A, Van Dyke DL: FISH analysis helps identify low level mosaicism in Ullrich-Turner syndrome patients.
