The IMMUNLITE 2000 DHEA-SO(4) is a solid-phase, chemiluminescent immunoassay. A polystyrene bead is coated with a polyclonal rabbit antibody specific for DHEA-SO(4). The patient sample and alkaline phosphatase-conjugated DHEA-SO(4) are added to the bead and incubated during this time, DHEA-SO(4) in the sample competes with the enzyme-labeled DHEA-SO(4) for a limited number of antibody binding on the bead. Unbound enzyme conjugate is then removed by washing, after which substrate is added. The chemiluminescent substrate, a phosphate ester of adamantyl dioxetane, undergoes hydrolysis in the presence of alkaline phosphatase to yield an unstable intermediate. The continuous production of this intermediate results in the sustained emission of light. The photon output, is inversely proportional to the concentration of DHEA-SO(4) in the sample.
