The IMMUNLITE 2000 DHEA-SO4 is a solid-phase, chemiluminescent immunoassay. A polystyrene bead is coated with a polyclonal rabbit antibody specific for DHEA-SO4. The patient sample and alkaline phosphatase-conjugated DHEA-SO4 are added to the bead and incubated. During this time, DHEA-SO4 in the sample competes with the enzyme-labeled DHEA-SO4 for a limited number of antibody binding sites on the bead. Unbound enzyme conjugate is then removed by washing, after which substrate is added. The chemiluminescent substrate, a phosphate ester of adamantyl dioxetane, undergoes hydrolysis in the presence of alkaline phosphatase to yield an unstable intermediate. The continuous production of this intermediate substance results in the sustained emission of light.
