The artificial substrate, 2-N-(hexadecanoyl) amino-4-nitrophenyl-phosphoryl-choline hydroxide, is cleaved by sphingomyelinase at pH 4.5 to yield the yellow sodium phenolate product plus phosphorylcholine.
