The purpose of this procedure is to amplify target segments of
DNA within the Class II region of the human major histocompatibility
complex such that allelic variability corresponding to the serologically
defined specificities for DR subregions can be identified. One of the
genomic regions common to most narcoleptic patients across all ethnic
groups is DQBeta1*0602. Setup includes mixing the HotStart Taq
polymerase chain reaction (PCR) mix with the DNA specimen and
dispensing the mixture in a thermoplate containing primers and thermal
cycling. After amplification, the PCR products are loaded onto an agarose
gel. After electrophoresis, the ethidium bromide stained gel is then
photographed and interpretation is made. (Moore SB: Unpublished
