Appropriately transported organisms are inoculated onto blood agar plates and into thioglycollate broth tubes, which are incubated anaerobically for 48 hours. Colonies are subcultured to determine aerotolerance, and obligately anaerobic organisms are identified by use of various differential media, biochemical tests, gas-liquid chromatographic analysis of volatile fatty acids produced during fermentation of glucose, and 16S ribosomal RNA gene sequencing.(Koneman EW, Allen SD, Janda WM, et al: Color Atlas and Textbook of Diagnostic Microbiology.
